But if we are to achieve it, we must be committed to do so as God commanded us, that means, we must follow God and live as Jesus lived and died. There is no other way out but God’s Way of life. The Kingdom of God can only be found in the relationship we create between God and us, and the way we continue to grow in this relationship. We can only have authentic peace in the long run if our lives are transformed and we are transformed by God. I want to tell you a story as I understand it. I cannot guarantee that what I am writing is absolutely true but as far as I can see, there is no way out of this dilemma. However, my story is a good story. This story depicts the good news. Landscape in T Cell Development {#S1}

During the last two decades, our understanding of the molecular machinery that instructs T cell development has significantly increased. This knowledge has been gained through the analysis of experimental models such as in the OT-I tumor model in which T cells bearing a specific TCR can be monitored over time ([@B1]) and through a combination of genetic studies and mathematical modeling ([@B2], [@B3]). In the OT-I model, most of the mutations that are acquired during the development of the T cells are targeting TCR genes. This suggests that during the development of T cells, the germline of the TCR can be maintained ([@B1]) and that there is a positive selection for a TCR that recognizes peptides presented by MHC molecules. Furthermore, the TCR repertoire is molded early in development through TCR rearrangement and selection for TCR structures that promote efficient signal transduction ([@B4]) and that are antagonistic to the ones potentially presented in infected and cancerous tissues ([@B5]–[@B8]). In addition, we now know that not only TCR-mediated signaling, but also the cytokine responsiveness and functions of the T cells require validation ([@B9]). This is also reflected by the stringent controls applied during T cell development. Here, both thymic- and peripheral-derived T cells differ in the binding capacity of their receptors. Specifically, the peripheral cells are often restricted to a small subset of the ligands recognized by the thymic repertoire, and once outside the thymus, such TCR–ligand pairs are not expressed by the cells ([@B10]). Consequently, after leaving the thymus, naïve T cells acquire the capacity to recognize thousands of self- and tumor-derived peptides in the periphery ([@B11]).
During the development, the cells of interest leave the thymus and migrate to the periphery, where they will reside most of their life time ([@B12], [@B13]). However, this education of the immune system has to be supported by balanced epigenetic reprogramming ([@B14]), in order to support robust T cell responses at the periphery.
